What is Tripterygium Wilfordii Extract Triptolide?
Tripterygium wilfordii is an endangered plant of the genus Tripterygium, which belongs to the family Celastraceae. In “Supplements for the Body,” it is stated to “open the duodenum and heal pain in the joints and bones.” Phytochemical analysis revealed that the dried leaves contained more than 100 compounds, including diterpenes, triterpenes, and alkaloids. One of the best characterized compounds was triptolide (molecular formula C20H24O6, CAS 38748-32-2).
Structurally, Tripterygium Wilfordii Extract Triptolide is a rosinan-type epoxy diterpene lactone consisting of three adjacent epoxy groups and an α,β-unsaturated five-membered lactone ring. These high-energy sections contribute to higher performance and power. The purity of the commercial product was ≥98% and appeared as a crystalline powder. They are soluble in DMSO and ethanol but are insoluble in water and should be stored at −20°C away from light.

What is triptolide used for?
• Rheumatoid arthritis (RA): A randomized controlled trial of more than 1,000 RA patients at 20 hospitals in China showed that oral triptolide (with a response rate of 0.2 to 0.3 ACR/tab) over 12 weeks reached 26%. The addition of methotrexate significantly reduced CRP and ESR.

What are the sources of triptolide?
Tripterygium Wilfordii Extract Triptolide contains 0.001%-0.008% (w/w) of triptolides, which means that 10–80 g of dry leaf extract can be selected per tonne. The procedure used 70% ethanol reflux extraction followed by macroporous resin formation followed by silica gel column chromatography followed by crystallization. This procedure can be scaled up to 100-gram volumes, where HPLC purity is ≥98%.
